The Department of Biology Sciences at the College of Education for Pure Sciences at the University of Basra organized a scientific lecture entitled (The effect of nanoparticles and natural chemical compounds on various cancerous and natural cell lines) by a master’s student (Ruqayah Kazem Jawad). The study included an overview of tumor emergence, its causes, treatment mechanisms and the new therapeutic mechanisms. Which can be used to eliminate cancer or to reduce the side effects of the treatments used, and one of the most important modern therapeutic mechanisms with a promising future is the Combination Therepy, as this type of treatment was addressed through an experiment combining both the nanomaterials of cerium oxide CeO2NP and ritonic acid RA, to test The extent of the synergism of the aforementioned substances on five of the cancerous and natural cell lines, and the necessary tests were conducted for each substance separately to identify its toxic potential, extract the half lethal concentrations IC50, and show the most important pathological and phenotypic changes using the double dye hematoxylin and eosin, as there were pictures Programmed cell death after staining of cell lines demonstrates treatment with both MDs T CeO2NP and RA separately and the two substances together with the acridine orange stain ethidium bromide, it was found that both substances have the ability to induce programmed cell death genes with both internal and external pathways in two types of cell lines CAL51 and HBL-100 after investigating three types of genes (Caspase8). , caspase9 p53) for apoptosis, using the simeQPCR technique.
Among the aims of the study:
1. Extraction of the lethal concentration IC50 of Ceo2NP and Ritonic acid nanoparticles based on the cellular activity of the above materials in the cell lines of normal and cancerous cells.
2. Investigate the nature of the relationship (synergistic or non-synergistic) between CeO2NP and R.A substances in natural and cancerous cell cultures, using the compsia program.
3. Determining the cellular pathways in which these materials operate in cell cultures when inhibition of normal and cancer cells occurs
It was also possible to reach the most important conclusions regarding the study, namely:
● Due to the toxicity shown by the nanostructured cerium oxide on both cancerous and natural lines, it is clear that it has no selectivity for cancer cells.
● RA shows clear toxicity on normal and cancerous streaks at high concentrations.
● We conclude that the toxicity of RA and CeO2NP are convergent in normal cell line HBL-100 while CeO2NP is more toxic than RA in carcinogenic lines (MCF-7, CAL51, AMJ31,).
● There is no antagonisem (antagonisem) between RA and CeO2NP in all tumor and normal lines, and it was concluded from the gene expression results that the action of the two substances alone is more than their combined action on the cell line HBL-100, while the combined action simulates one of the substances in the cancer cell lines. .
● RA induces programmed cell death in cancer cell lines by a pathway that differs from inducing programmed cell death in normal lines.
